Addition of amino acid analogues into the set of building blocks for protein synthesis has expanded the scope of protein engineering. The two methods, site-specific incorporation using one of the stop codons and residue-specific incorporation by reassigning codons for one amino acid, have been developed for expressing recombinant proteins including unnatural amino acids in vivo. However, each method has suffered from limitations such as a low expression yield of the site-specific method for proteins with multiple stop codons, and the need to sacrifice one amino acid to incorporate one unnatural amino acid using the residue-specific method. In this presentation, we will introduce our current effort to develop a new method to incorporate amino acid analogues into protein by reassigning a specific codon for unnatural amino acids. This system overcomes the limitations of current methods and, we expect, will provide a powerful tool for engineering proteins with novel compositions. |
