Oral delivery of gene loaded Protamine-TCA nanocomplexes to treat hepatic cancer |

Gene delivery has gained significant importance in cancer therapy due to its efficiency and minimal toxicity. However, oral delivery of therapeutic genes remains a challenge due to gastrointestinal barriers and non-specific targeting. Here we report the synthesis of Protamine-TCA loaded genecomplexes to treat hepatic cancer. Cationic protein, protamine helped in the conjugating the negatively charged gene. The TCA helped preventing the complexes from degradations. NMR studies confirmed the conjugation of TCA to protamine. The gel electroporation, DLS and Zeta potential analyses confirmed the formation of the gene complexes. Serum stability studies further confirmed the stability of the formed polyplexes. In vitro toxicity studies showed negligible toxicity profiles of the carrier. Moreover, studies on Caco-2 and MDCK cell lines confirmed the efficient gene expression from the protamine-TCA gene complexes. We believe that our non-viral vectors could be efficient for oral gene delivery. |
