제목 Dendritic Hybrid Polymer as a Novel Carrier of Bioactive Agents
초록 Dendritic hybrid consists of a linear polymer and dendritic units chemically conjugated at the end of each chain of linear polymers. It is expected that the dendritic hybrid shows a tendency to form a unimolecular micelle and has a precise control over the size by changing the degree of polymerization of the linear polymer part as well as the generation number of dendritic part. Unimolecular micelle leads to the increased stability in the blood stream compared with micelles due to the absence of critical micelle concentration. A novel dendritic hybrid polymer was prepared with biocompatible and biodegradable 4-arm star poly(ε-caprolacton) (PCL) as a hydrophobic internal core and poly(ethylene glycol) (PEG) as a hydrophilic dendrons. In addition, a model system of the synthesized polymer in a selective solvent is simulated by a dynamic Monte Carlo method to find a polymer architecture optimized for drug loading. Static properties such as the chain size and the loading capacity of drug are calculated as a function of various architectural parameters such as the generation number of a dendron. It is found from both experiment and simulation that the polymer structure contains a holey region between the inner core of collapsed star blocks and the outer shell of bulky dendrons, suggesting that the drugs can be easily encapsulated in the holey region sandwiched between two dense regions of the core and the shell.
