Self-assembled Y-RNA structure of CRISPR/CAS9 for a multifunctional genome editing |

The RNA-guided nuclease CRISPR (clustered regularly interspaced short palindromic repeat)/CRISPR-associated (Cas) protein 9 system is a new emerging gene editing approach in the field of gene therapy. In this study, we prepared the self-assembled Y-RNA nanostructures as multifunctional guide RNAs (gRNA) for Cas9 delivery. To prepare each arm of Y-RNA structures, we utilized the rolling circle amplification of DNA template. Amplified RNA products were treated with RNase H and the length of gRNA was carefully controlled by site-specific cleavage. After complexation of Cas9 with gRNA, three distinct Cas9/gRNA complexes can self-assemble to form Y- RNA structures for region-specific (loco-specific) gene editing in mammalian cells. Efficient and programmable gene editing has been demonstrated in MEF cells, which co-express the green (G), red (R), and blue (B) fluorescence proteins. |
