Bioorthogonal functionalization of genome editing protein by amino acid tagging and click chemistry |

Recent advances in genome editing technologies based on the CRISPR-Cas system has enabled rapid and accurate alterations of genomic information in various organisms. Functionalization of the genome editing protein Cas9 with various bioactive molecules allows diverse applications of the CRISPR system for both medical and industrial purposes. Herein, we report a versatile approach for introducing chemoselective functionalities into the Cas9 protein via biorthogonal noncannonical amino acid tagging. We used L-azidohomoalanine (AHA) to replace the methionine groups in Cas9, which can be tagged via azide-alkyne cycloaddition for further bioorthogonal functionalization. We characterized the functionalized protein by labeling with fluorophores and visualization. This approach will widen the applications of CRISPR, especially for therapeutic purposes where genetic modification should be performed through a nonviral procedure. |
