Gene Cloning and Characterization Involved in the Biosynthesis of Dihydrochalcomycin in Streptomyces sp. KCTC 0041BP |

The 63 kb region in this fragment was found to contain 25 ORFs. The products of all of these ORFs play a role in dihydrochalcomycin biosynthesis and self-resistance against the compounds synthesized. The 39.6 kb PKS was also found. The genes responsible for deoxysugars were found. To confirm the involvement of this gene cluster in dihydrochalcomycin biosynthesis disruption of dehydratase (DH) domain in module 3 was carried out. The disruption was characterized by southern hybridization. The loss of dihydrochalcomycn production was confirmed by LC-MS and antibacterial susceptibility tests. The disruption indicated that the cloned gene cluster is responsible for dihydrochalcomycin production. |
