Enhanced myelination of pluripotent stem cell-derived oligodendrocytes on extracellular matrix-modified nanofibrous scaffolds |

Previously there have been few studies to enhance myelination process of stem cells via tissue-engineering strategies. In this study, we developed a functional culture platform which is composed of extracellular matrix (ECM) and electrospun nanofiber scaffolds to facilitate myelination process between human iPSC-derived oligodendrocytes and induced neuronal cells. Our results indicated that ECM-functionalized nanofibrous scaffolds efficiently guided myelination of hiPSC-derived oligodendrocytes co-cultured with iN cells by promoting cellular alignment and interactions along the nanostructures. In conclusion, we suggest that in vivo-like ECM-modified scaffolds can efficiently provide a functional myelination platform, which has huge potential to apply for demyelination disease modeling and regenerative medicine for the treatment of demyelination diseases.
