Mass spectrometry approaches for comparative analysis of N-linked glycan from Wild-Type and α1,3-Galactosyltransferase (GalT) Gene Knock-Out Pig Fibroblasts |

Carbohydrate antigens expressed in porcine cells are known as major barriers to pig-to-human xenotransplantation, but there is no extensive study analyzing N-glycans expressed on the GalT-KO porcine tissues or cells. Thus, we performed MALDI-based relative analysis of totally N-glycans from wild-type (WT) and GalT-KO pig fibroblasts. As a result, we identified and quantified a total of 47 N-glycans. First, the absence of galactose-alpha-1,3-galactose (α-Gal) residues in GalT-KO pig cells was confirmed. Additionally, we identified that the the level of total fucosylated N-glycans from GalT-KO porcine fibroblasts is much higher than from WT porcine fibroblasts and the comparative quantity of the N-glycolylneuraminic acid (NeuGc) antigen is slightly higher in the GalT-KO pigs. Thus, this study could aid a better understanding of the changes of cellular glycan on GalT-KO pig for successful xenotransplantation. |
