Polyacrylamide gel stabilization electrophoresis : Alpha-helix stabilization via copolymerization |

Our work reports on the development of a novel strategy to stabilize bioactive alpha-helical peptides into a polyacrylamide hydrogel network. We synthesized an RNA binding peptide derived from the human immunodeficiency virus type-1 (HIV-1) Rev protein and two acryloyl groups were added to the peptide. We named it "bis-Rev". We prepared gels for electrophoresis in TBE buffer at 4 ℃. We confirmed that the bis-Rev are covalently captured in polyacrylamide gel by comparing acryloylated or non-acryloylated peptides on gel electrophoresis. Additionally, RRE mutant was used to confirm the specificity of the RNA-peptide binding. In conclusion, we developed a novel method for stabilizing alpha-helical peptides by copolymerization within a polyacrylamide gel. |
