Disulfide Cross-linked Heparin Nanogels for Inducing Apoptosis |

Heparin, a highly sulfated glycosaminoglycan, was recently shown to induce apoptotic death of cancer cells by interfering activation of transcription factors. Here we report disulfide cross-linked heparin nanogels for efficient intracellular delivery of heparin. These nanogels were produced through formation of disulfide bonds between thiolated heparin molecules that were solubilized by forming nanocomplexes with poly(ethylene glycol) in organic solvent. The resultant heparin nanogels had a very stable structure with an average diameter of 181.3±9.4 nm in aqueous solution. The disulfide cross-links were cleaved in intracellular reductive environment. The heparin nanogels significantly inhibit the proliferation of mouse melanoma cells compared to unmodified heparin. Cellular uptake and apoptotic effect of these nanogels were evaluated by confocal microscopy and flow cytometric analysis. |
