Bioreducible Polymer and Graphene Oxide for Enhancing Chondrogenic Differentiation and Therapeutic Angiogenesis of Adult Stem Cells |

Graphene oxide (GO) can be utilized as both a cell-adhesion substrate and a growth factor protein-delivery carrier for the chondrogenic differentiation of human adult stem cells (hASCs). Conventionally, chondrogenic differentiation of stem cells is achieved by culturing cells in pellets and adding the TGF-β3 to the culture medium. We utilized GO sheets to adsorb fibronectin (FN) and TGF-β3 and were then incorporated in pellets of hASCs. The hybrid pellets of hASC-GO enhanced the chondrogenic differentiation of hASCs by adding the cell-FN interaction and supplying TGF-β3. We also developed a branched poly(disulfide amine) (B-PDA) for intracellular delivery of Fas siRNA into hASCs. The Fas-silenced hASCs can then be formulated as enlarged stem cell spheroids and transplanted into mouse ischemic hind limbs as paracrine secretors for therapeutic angiogenesis. These methods may provide a new platform for stem cell application for regenerative medicine. |
