Synthesis of Core Cross-linked Polymeric Micelles by Diels-Alder Click Reaction for Drug Delivery |

Well-defined poly(ethylene glycol)-b-poly(furfuryl methacrylate) (PEG-b-PFMA) block copolymers were prepared by the reversible addition-fragmentation chain transfer (RAFT) polymerization. In water, the copolymer formed micelles with PFMA in the core and PEG in the shell. The hydrophobic core domain was used for the incorporation of doxorubicin (DOX) anticancer drug and crosslinking by Diels-Alder using disulfide-containing cross-linker at 60oC. The core cross-linked micelles formed spherical structure with average diameter 68 ± 4 nm. In vitro drug release showed that DOX release from micelles was only 19% at normal physiological conditions, whereas 52% DOX was released in reductive acidic environment. The results indicated that Diels-Alder click reaction could serve as an effective cross-linking method for preparation drug delivery system. |
