Preparation and Characterization of A Novel Aptamer Biosensor Gate |

A novel aptamer biosensor gate was prepared to detect the change of electronic and optical signals during and after the binding of aptamers on the chip to biomolecules. A silicon wafer pre-coated with gold (Au-Si wafer) was used as a model substrate and anti-thrombin aptamer, which specifically recognized thrombin in the blood, was used as a model aptamer with a sequence of d(GGTTGGTGTGGTTGG). A solution of thiol modified aptamers was coated on Au-Si wafer. The chip was washed and then observed with an atomic force microscope (AFM) and a scanning electron microscope (SEM), which confirmed the successful binding of anti-thrombin aptamers on the Au-Si wafer. The binding capacity of the aptamer biosensor to FITC labeled thrombin will be discussed in detail. The novel aptamer biosensor gate will be further applied for the development of aptamer-nanowire biosensors. |
