Bioorthogonal functionalization of Cas9 protein with bioactive molecule via click chemistry |

Recent advances in genome editing technologies based on the CRISPR-Cas9 are enabling the rapid and accurate alterations of genomic information in various organisms. The genome editing protein Cas9 can be functionalized with many different bioactive molecules and this allows various application of CRISPR system for both industrial and medical purposes. Herein, we introduce chemoselective functionalities into the Cas9 protein via biorthogonal noncannonical amino acid tagging. We used azidohomoalanine (AHA) to substitute methionine groups of Cas9 and this can further be tagged by azide-alkyne cycloaddition with dibenzocyclooctyne. The high reaction rate and its desired biocompatibility allows the functionalized Cas9 for therapeutic purposes through a nonviral procedure. This approach will have great potential for creating bioorthogonally functionalized Cas9 which can widen the application of CRISPR, especially for the development of therapeutic proteins with new and improved properties. |
