L-Tyrosine is an important compound because of one of essential amino acids and precursors to pharmaceutical drugs and valuable chemicals. In E. coli, L-Tyrosine is naturally produced from glucose via enzymes in glycolysis and enzymes for conversion of erythrose-4-phosphate (E4P) and phospho-enol-pyruvate (PEP) to tyrosine. However, in order to efficiently produce L-Tyrosine, it is required to optimize biosynthetic pathway for L-Tyrosine production. Here, we designed biosynthetic pathway for tyrosine production in E. coli chromosome to sufficiently enhance metabolic flux to tyrosine using synthetic promoter and synthetic UTR and then we regulated expression level of enzymes related with PEP to find the optimal metabolic flux between cell growth and tyrosine production. As a result, engineered strain showed significant increase in tyrosine production compared to wild-type strain. |
