Over the past years, various nano-carrier systems, such as micelles, liposomes, nanoparticles, and polymersomes, have been extensively investigated for efficient delivery of therapeutic drugs and genes. Among various platforms, polymer micelles that are constructed by self-assembly of amphiphilic polymers in aqueous media have shown excellent performance due to their various advantages, such as unique core-shell structure, long circulation time, and EPR effect. In this study, we synthesized biarmed methoxy poly(ethylene glycol) (mPEG)-dexamethasone (Dex) conjugates mPEG-Dex as a bioactivatable nuclear targeting system. The conjugates were investigated with regard to their physicochemical characteristics, self-assembling behaviors, drug loading capacity, and in vitro efficacy in tumor cell culture systems. The bioreducible activation mechanism of mPEG-Dex micelles is assumed to induce nuclear localization of doxorubicine (DOX) and thus enhance therapeutic efficacy. |
