Preparation and Characterization of Double-Layered PLGA Microsphere for Effective Controlled Release of Raloxifene HCl |

Raloxifene HCl is a selective estrogen recepter modulator(SERM) and is currently being used for prevention of osteoporosis in postmenopausal women. We prepared raloxifene HCl loaded double-layered PLGA microspheres using O/O/W double emulsion solvent evaporation method. The raloxifene HCl loaded double-layered PLGA microsphere was characterized on the surface, and the cross-section morphology by SEM. In vitro release behavior of raloxifene HCl was analyzed by HPLC over 4 weeks. Double-layered PLGA microspheres showed smooth surface and obvious difference core and outer-shell by SEM observation. The influences of preparation parameters, such as molecular weight of PLGA and initial drug loading ratio, have been observed in the release behavior of raloxifene HCl. These results showed that the release behaviors can be effectively controlled by the preparation parameters. |
