제목 HPMA copolymer-cyclic RGD Conjugates for Imaging and Targeted Drug Delivery to Sites of Tumor Angiogenesis
초록 Cyclic arginine-glycine-aspartic acid (RGD) peptides bind with high affinity to αvβ3integrins overexpressed on angiogenic blood vessels. N-(2-hydroxypropyl)methacrylamide (HPMA) copolymers are well-characterized water-soluble polymers used for targeted drug delivery to solid tumors. We have synthesized HPMA copolymers with pendant cyclic RGD4C and RGDfKin the side chains to deliver radionuclides and drugs to sites of tumor angiogenesis (DB Pike & HGhandehari, Adv Drug Deliv Rev, 62:167, 2010). Such conjugates were localized in tumor xenografts of prostate, lung, and breast to a higher extent compared to passive delivery and enhanced contrast of magnetic resonance and scintigraphic imaging agents.The localization of 90Yittrium, 17-(6-aminohexylamino)-17-demethoxygeldanamycin, and docetaxel in prostate tumors was increased when delivered by the polymer-peptide conjugates. These conjugates show promise as multifunctional theranostic platforms for imaging and therapy of solid tumors.
