Nitric Oxide-Releasing Tethered Lipid Bilayer Membrane |

The controlled release of nitric oxide (NO) could be considered to be one of the most promising approaches for preventing restenosis and thrombosis. The aim of this study is to investigate the use of tethered supported lipid bilayer (TLB) coating on stent with a lipophilic NO donor incorporated within the phospholipid bilayer, to achieve a NO controlled and sustained release rate. The NO releasing TLB membranes are prepared via two steps 1) tethering phospholipids on a metal surface and 2) fusing NO donor containing liposomes with the lipids pre-tethered on the surface. Surface characterizations including confocal microscopy and atomic force microscopy (AFM) showed successful bilayer formation on the surface of titanium. NO release profiles were sustained up to 24 hrs. In vitro cell study demonstrated that NO release enhanced endothelial cell proliferation. Finally, these results open a door for the potential use of TLB coating that utilizes therapeutic agents in biomedical devices. |
