With the knowledge that nitric oxide (NO) serves as a therapeutic agent, we aimed to investigate into strategies to efficiently store and controllably deliver NO to the site of interest to achieve new successful clinical applications. Here, we present an on-demand controlled NO delivery system based on IMesNO, the N-heterocyclic carbene-based NO donor (NHCNO). Unlike other typical NO donors such as N-diazeniumdiolates (NONOates) and S-nitrosothiols (RSNOs), IMesNO stably preserves NO, but rapidly releases NO on-demand upon irradiation with high-intesity focused ultrasound (HIFU). Futhermore, for its potential bioapplication, the IMesNO-loaded polymer micelles (IMesNO@MCs) are designed to preferentially accumulate in a targeted tumor and respond to HIFU irradiation for simuli-responsive NO release, leading to site-specific and controllable NO release in vivo. |
