Nitric Oxide (NO) is an endogenously synthesized small molecule that regulates multiple functions in physiological processes, such as the control of vasodilation, immune responses, the inhibition of platelet adhesion and aggregation, angiogenesis, apoptosis and wound healing. Various studies have been implemented for releasing and delivering nitric oxide. However, small molecule-NO donors have short half life time and initial burst releasing profile. Herein, we report feasibility of mPEG-PLGA copolymer as nitric oxide releasing nano-polymersomes. mPEG-PLGA copolymer was synthesized by ring-opening polymerization of lactide, glycolide and mPEG as a macro-initiator. Double emulsion method was used to prepare mPEG-PLGA incorporating NONOate(DETA/NO). The nano-polymersomes with different molecular weight of PEG released nitric oxide different periods and didn’t show cytotoxicity in the various cell types, including fibroblasts, HUVEC and epithelial cells. |
