제목 | Solid-phase synthesis of thiol compounds as a spacer for DNA chip |
초록 | In the development of biochip system, modification of a chip surface for biomolecules immobilization is a key step and should be reliable and sensitive. We have developed a new synthetic strategy for the synthesis of spacers to immobilize biomolecules on the chip surface. We then demonstrate the reliability of surface modification by detection of DNA molecules on the chip. Synthetic strategy for the spacer is based on the solid-phase protocol by using 2-chlorotrityl chloride resin. Solid-phase synthesis can give us many advantages in the purification and characterization of the spacer molecules compare to the solution-phase synthesis or on-chip synthesis. The linkers contain triethylene glycol unit (TEG) in order to increase reliability and sensitivity of the DNA chips. We applied the spacers to a gold substrate for electrochemical DNA detection. We confirmed each reaction on the resin by FT-IR and elementary analysis. The surface modifications were characterized by contact angle goniometry, atomic force microscopy, X-ray photoelectron spectroscopy. We prepared DNA arrays on the gold substrate, and detected single base mismatched DNA by cyclic voltammetry (CV), and demonstrated the utility of our sensor system. |
