Synthesis of Novel Dendritic Photosensitizers for Photodynamic Therapy |

Recently, we have reported ionic dendrimer porphyrin as an efficient photosensitizer for photodynamic therapy. Although dendrimer porphyrin micelle exhibited significant phototoxicity to tumor cells, its application field is limited due to the low tissue penetration ratio of excitable light. Therefore, different types of poly(benzyl ether) dendrimers, were synthesized and well characterized as potential candidates of photosensitizer. These dendrimers exhibited large absorption bands at quite longer wavelength region than that of previously reported dendrimer porphyrin, and formed uniform micelle when the dendrimers mixed with same charge ratio of PEG-PLL block copolymer in 10 mM of PBS. Oxygen consumption of each dendrimers and micelles in 10 mM PBS were measured upon the light irradiation. As a result, partial pressure of oxygen in the phthalocyanine dendrimer solution was significantly reduced under the irradiation of 680 nm laser light. The evaluation of in vitro phototoxicity to the hela cells revealed that the phthalocyanine dendrimer polyion complex micelle is an efficient photosensitizer for photodynamic therapy with long wavelength light absorption. |
