Transplantation of ex vivo cultured cells is one of the most emerging research fields in regenerative medicine. One innovative technique involves poly(N-isopropylamide)(PNIPAAm)-grafted surface, in which confluent cell sheet is harvested and transplanted to target tissue, demonstrating improved engraftment efficiency and cell viability. However, the floated cell sheet should be translocated to intermediate surface, which may result in abnormality of cell membrane integrity, and may require relatively long processing time. Given that, we developed cell-interactive and temperature responsive hydrogels as a novel substrate to culture confluent cell layers, which could translocate cell sheet to various targets by simple one-step stamping process. This presentation summarizes our progress in this approach including characterization of hydrogels, in vitro cell stamping processes, and its therapeutic applications. |
