제목 In Vitro and In Vivo Antitumor Activities of BCNU-loaded PLGA Wafer against L 1210 Mouse Leukemia
초록 The localized, controlled delivery of anticancer agents using biodegradable polymeric implant is an alternative to the systemic administration of chemotherapeutic agents for treating malignant brain tumors. 1,3-bis(2-chloroethyl)-1-nitrosourea(BCNU) is one of the chemotherapeutic agents used for brain tumors, however, it's short half-life and systemic toxicity. In this study, BCNU-loaded PLGA disks were prepared as a new formulation for the controlled release of BCNU and their antitumor activity was evaluated in vitro and in vivo. In anititumor acitivity in vitro, change of cell density by BCNU released from wafer was assayed by dye exclusion method. In antitumor activity in vivo, a solid tumor was induced by the s.c. injection of 0.1 ml of L 1210 cell suspension (5 X 104 cells per mouse) in the flanks of BDF1 mice. Treatment was initiated at day 7 after injection of cells. Small incision was made above the tumor and BCNU-loaded PLGA wafer was implanted next to the tumor. Antitumor activities of the groups treated with BCNU-loaded PLGA wafers were evaluated by tumor size reduction and median survival rate.
