제목 | Bioavailability and Pharmaceutical Formation of Drug Nanoparticles of α-Lipoic Acid having Anti-Obesity and Appetite Suppression |
초록 | The drug delivery systems of α-lipoic acid (ALA) which stands in the spotlight of the novel anti-obesity drug were investigated. For a long time, ALA has been recognized as the strongest antioxidant in that it has partial solubility in both water and oil. Furthermore, most researchers agree that its stability in the body is the best in hundreds of anti-oxidants and ALA does not especially have any side effects. The research about the useful quality of ALA was more progressed because a small amount of ALA is produced from biosynthesis in the body [1]. Recent results had proved that ALA has the effects of not only anti-obesity but also appetite suppression when an overdose of ALA is taken [2]. Pharmaceutical size reduction methods such as ball-milling, micro-emulsion, etc., was applied for the production of pharmaceutical dosage forms of ALA which release the active ingredient more rapidly and have greater bioavailability than previous ordinary dosage forms. Nevertheless, because of the chemical characteristics (S-S bond: disulfide linkage) of ALA, it is easy to undergo crosslinking reactions by heat, light, ions, or friction heat generated by mechanical energy. This is a serious problem in the pharmaceutical formulation of ALA making the preparation window of ALA relatively narrow. With special conditions, micro-emulsion and nano-comminution were selected as the basic experiment to relieve these difficulties. The drug nanoparticles of ALA produced by these methods was administered to standard rats and showed more effective decline of food intake than microparticle ALA.
