The CRISPR-Cas9 system is a powerful sequence-specific gene editing tool. In previous studies, researchers had to undergo trials and errors to make different silencing efficacy using the CRISPR interference system, since they had to create and experiment with multiple sgRNAs. In this study, the correlation between hybridization energy of the sgRNA-target sequence complex and repression level of the targeted gene in CRISPR interference system was investigated to predict the silencing efficacy. We generated sgRNA library hybridizing with a target gene at various strengths and used the library to create target gene library that silenced at various levels. The more mismatches between sgRNA and target sequence were inserted, the more decrease was found in the repression level of the targeted gene (R2 value is 0.64). This indicates that we can successfully predict how much a targeted gene will be repressed when a particular sgRNA is designed. |
