제목 | Sol-Gel-Sol Transition of MPEG-PCL Diblock Copolymers |
초록 | Polymers that response to external stimuli have been investigated for the controlled-release drug carriers. Especially, PEG-based diblock and triblock polyester copolymers, stimulating to temperature were studied as injectable drug delivery system because of their nontoxicity, biocompatibility and biodegradability. We synthesized the diblock copolymer consisting of methoxy polyethylene glycol (Mn=750 g/mole) and ε-caprolactone by ring opening polymerization in the presence of HCl․Et2O at room temperature. The concentrated solution of synthesized diblock copolymer represented a sol to gel phase transition as the temperature increased from room temperature to body temperature. Above 45 oC, these block copolymers displayed a sol phase. To confirm the gel formation at body temperature, we observed the formation of gel in the mice body after injection of 25 wt% aqueous solution of each block copolymer. After 1 month, we investigated the gelation without dispersion in mice. The results obtained in this study confirmed the feasibility as biomaterials of injectable implantation for controlled release of drug and protein delivery.
