제목 Effect of SIS Content of Proliferation and Phenotype for Intervertebral Disc Cells on PLGA/SIS Scaffold
초록 Annulus fibrosus(AF) and nucleus pulposus(NP) cells were seeded in PLGA and PLGA/SIS scaffold. Cellular viability and proliferation were assayed by cell counting. Reverse transcription polymerase chain reaction(RT-PCR) was assessed to measure mRNA expression for Type I collagen and Type II collagen of IVD cells in scaffolds. SEM observation of disc cell population expanded on scaffold. After seeded cell, we observed charcterization of disc cell by cell counting and RT-PCR on 1, 2, and 3 day. In counting result, we observed that 10 wt% SIS content scaffold better than only PLGA scaffold for cell proliferation. RT-PCR and SEM results, we confirmed more express Type I collagen and type Ⅱ collgen in PLGA/SIS scaffold. when we cultured disc cell in vitro, we observed better to keep the characteristic of disc cell in contents of SIS.
