Cell membranes are important constituents to keep homeostasis in the biological system. Model cell membranes in the form of planar membranes or liposomes can provide an excellent platform for biophysical studies of cell behaviors at the molecular level. In this work, we studied the interactions between ionic liquids and a biomimetic membrane with a model membrane protein, gramicidin A. To elucidate the membrane behaviors, we used two model systems: fluorescence quenching assay using liposomes and biophysical assay using a planar membrane. As a result, ILs incorporated in to the model membranes increased lifetime of the ion channels via stabilizing membrane deformation and decreased the permeability of the ions by changing the surface charge of the membranes. In addition, to prove physical and electrical phenomena of the experiment, we further conducted MD simulation. Our results shown here will provide provide a useful tool to design more biocompatible ILs and to analyze toxicity of ILs or other small molecules. |
