PNIPAAm with the controlled molecular weights was synthesized by the atom transfer radical polymerization of NIPAAm using t-butyl 2-chloropropanoate, and treated with trifluoroacetic acid to give carboxyl-terminated PNIPAAm. Artery wall binding peptide (AWBP; Cys-Gly-Arg-Ala-Leu-Val-Asp-Thr-Leu-Lys-Phe-Val-Thr-Gln-Ala-Glu-Gly-Ala-Lys), a specific targeting peptide, was conjugated to the carboxyl-terminated PNIPAAm to enhance the drug transfer to artery wall cells. 1H-NMR and GPC analysis confirmed the composition and molecular weight of the obtained polymer and indicated that AWBP was successfully incorporated into PNIPAAm. AWBP-b-PNIPAAm, which is targeting artery wall, offers the possibility of intelligent drug capsule communicating with cells to release its pharmacological activity. |
