제목 Fabrication of BMP2-PVA-Gelatin Hydrogel Loaded HAp Scaffold for Bone Tissue Engineering.
초록 In this experiment, we fabricated PVA-Gelatin hydrogel loaded HAp Scaffold by Sponge Replica method with Bone Morphogenetic Protein 2 (BMP2) loaded within the hydrogel matrix. Porous HAp saffold was fabricated using 45 ppi Polyurethane sponge. After several times of dip coating and subsquent drying, samples were burn out and sintered by microwave furnace at 1300°C. PVA-Gel hydrogel was prepared by disolving PVA&Gel in hot water and then cooling to room temperature. Then HAp Scaffold was loaded with Hydrogel. X-ray diffraction analysis, fourier transform infrared spectroscopy (FT-IR) was performed to characterize the phase identity. Microstructures of the composites were observed using scanning electron microscopy and compressive strength, porosity was also measured. In vitro F-actin staining, dead/live cell staining, cyto toxicity and, proliferation were carried out to investigate in detail about the bio-compatibility of the system. In in vivo experiment, implanted hydrogel loaded HAp scaffold was collected after 1 and 2 months of implantation and new bone formation was evaluated by using micro-CT analysis and histological staining.
