Fabrication of artificial scaffolds imitating the porous network of native bone and its in vitro and in vivo evaluation |

A novel artificial scaffolds with hydroxyapatite (HAp) was designed and fabricated to imitate native bone with a multi-pass extrusion process. The conceptual design contained unidirectional multimodal channels that mimicked typical long bone architecture, and a central canal, which houses the bone marrow and micro porous preforms at the exterior part of the scaffold to facilitate rapid bio mineralization. External and internal fissures were minimized during microwave sintering at 1100°C. Detailed microstructure and mechanical properties of the fabricated artificial bone were examined by SEM, XRD and mechanical measurement as compressive strength. The initial biocompatibility was examined for cell proliferation of MG63 osteoblast-like cells using MTT assay, and cell adhesion was visualized by CLSM. In vivo implantation after 4 weeks and 8 weeks showed integration of the scaffold to the native tissue and formation of bone tissue within the pore network was confirmed with micro-CT analysis and histological staining. |
