The systematic combination of nano-hydroxyapatite (n-HAp), BMP-2, and porous polymer scaffolds is a significant challenge for high-performance bone regenerative materials. Herein, we developed n-HAp-immobilized porous poly(lactide-co-glycolide) (PLGA) microspheres that can induce the sustained release of BMP-2. The surface-exposed n-HAp can display its inherent osteoconductivity of 100%, and the BMP-2 release can induce stem cell differentiation to osteoblasts. Real-time PCR and histological analyses including H&E, Masson’s trichrome, and alizarin red S staining showed that the incorporation of BMP releasing property contributed to the enhanced formation of bone tissues by n-HAp-immobilized PLGA microspheres. The rational combination of n-HAp, BMP-2, and porous PLGA microspheres can generate the new type of injectable scaffolds for bone tissue engineering. |
