제목 Endolytic Folic acid (FA) derivatives for effective intracellular delivery of siRNA
초록 Folic acid is one of active targeting ligands often used for drug delivery systems. Folic receptors (FR) are abundantly expressed on many types of cancer and their strong binding affinity with folic acid make them very attractive targeting system for delivery of genetic materials such as pDNA and siRNA. However, many studies have reported that FA-siRNA conjugates are not effective, when delivered alone, without employing endosomal releasing agents. In this study, we have synthesized various endolytic FA derivatives which can facilitate the intracellular delivery of siRNA as well as enhancing the endosomal release of siRNA into the cytoplasm. Targeting and effective gene silencing have been verified in GFP expressing KB cells.
