제목 A Novel Hyaluronic acid - Peptide Conjugate by Michael Addition for Formyl Peptide Receptor Like - 1 Receptor
초록 Hyaluronic acid (HA) - peptide (WRYMVm) conjugate for formyl peptide receptor like - 1 (FPRL-1) receptor was successfully synthesized. Aminoethyl methacrylated HA (HA-AEMA) was synthesized and conjugated to C-WRYMVm via Michael addition reaction between metharylate groups and thiol groups in cysteine. HA-peptide conjugate was purified by fractionation using gel permeation chromatography (GPC). The formation of HA-peptide conjugate could be confirmed by GPC and 1H nuclear magnetic resonance (NMR) analysis. The peptide content in HA-peptide conjugate was quantified by measuring the absorbance at 280 nm with GPC. The biological activity of HA-peptide conjugate was confirmed by in vitro RBL-2H3 cell activity tests measuring the elevation level of phospho-extracellular signal-regulated kinase (pERK). The novel bioconjugation of HA-AEMA to peptide molecule with thiol groups might greatly contribute to elongate its half-life, which can be successfully applied for various biopharmaceuticals.
