PLGA has been widely used as medical applications. However, the decomposition products of the PLGA are known to acidify surrounding tissue of the implanted site, thus causing an inflammatory reaction. In this study, we developed the PLGA/β-TCP/Mg(OH)2 that induces anti-inflammatory and osteoconductive effect. The inorganic compounds were uniformly blended in melted PLGA polymer matrix via a twin extrusion process. The thermal and mechanical properties were characterized by using GPC, TGA, and UTM. The fracture surface morphology of nanocomposites was observed by FE-SEM. The anti-inflammatory activity were evaluated by ELISA assay for IL-6 and TNF-α, respectively. The osteoclast differentiation was evaluated by TRAP-staining analysis. The osteoblast differentiation was evaluated by PCR, VK, ARS staining analysis, respectively. As a result, PLGA/ β-TCP/Mg(OH)2 developed in this study are expected to be applicable to orthopedics as well as various bio-implantable materials. |
