제목 ECM hydrogel loaded 3D printed polycaprolactone scaffold for enhanced bone regeneration
초록 Numerous bone fractures and bone defects occur due to accidents or other sources. In case of depressed skull fracture, fracture sites need scaffolds to act as a buffer for variable brain pressure. In order to meet these requirements, there is a huge demand for the fabrication of scaffold in a specific form. Recently, 3D printing technology is gaining attention among researchers for biomedical applications. In addition, Extra Cellular Matrix (ECM) contains growth factors and supports for cell attachment. Thus, in this study, we designed ECM hydrogel loaded 3D printed polycaprolactone (PCL) scaffold for bone regeneration. ECM was first prepared with de-cellurized porcine skin, then ECM hydrogels were fabricated by enzymatic digestion. PCL scaffold was printed by fused deposition modeling (FDM) method and surface modified by 1M NaOH. The prepared ECM hydrogels were loaded finally in a 3D printed PCL scaffold. The morphology, in-vitro biocompatibility of ECM loaded PCL were evaluated and compared with PCL scaffold. In-vivo performance of the composite scaffold was further tested using rat model.
