제목 Tumor-Homing, Size-Tunable Clusterized Nanoparticles for Anticancer Therapeutics
초록 We present herein a pH-responsive dynamic nanostructure based on gold nanoparticles with DNA assembly and evaluate its working as a potential drug delivery agent with tumor specific accumulation. Clusterization of gold nanoparticles induced by hybridization of each DNA sequence via i-motif DNA provides higher tumor targeting as well as drug loading capability. After cellular uptake, drug was released by disassembly of nanoparticle cluster into single nanoparticles with respect to pH decrease at the late endosome. Furthermore, the expression of anti-apoptotic Bcl-2 protein was down-regulated by antisense-modified gold nanoparticles, thus drug-mediated apoptosis was significantly accelerated by sensitizing the cancer cell toward drugs. Our size-tunable clusterized nucleic acid grafted gold nanoparticles provide tumor-homing property during blood circulation and verified its potential as a multifunctional therapeutic agent in vivo as well as in vitro.
