Cytochrome P450 monooxygenase is a well-known biocatalyst for broad substrates range, activation molecular oxygen under normal pressure & room temperature, and highly stereo- and regio-selective on complex compounds. As use of the cytochrome P450 as promising industrial catalysts, a whole-cell biotransformation system should be constructed. In this study, confronting problems related to P450 catalyzed biotransformation would be introduced. As a model system, CYP153A, a well-known ω-hydroxylase, is used to produce hydroxy long-chain fatty acids. Especially, ω-hydroxy long-chain fatty acid is one of valuable chemicals as a key component of cosmetic industry related to pseudo-ceramide synthesis. The aim of this study is to demonstrate current hurdles for biotransformation of hydroxy long-chain fatty acid and discuss breakthrough. |
