제목 In Vivo Cytotoxicity and Biodistribution of Cationic Emulsion-Mediated Gene Delivery Systems as Non-Viral Vectors
초록 Non-viral gene delivery system is attractive due to low toxicity and reproductive transgene expression compared to viral vector commonly used in gene therapy protocols. We developed sufficiently stable and biocompatible emulsion complex, which was positively charged and spherical. This study evaluated the safety and the gene transfection ability of the complex in vitro and in vivo by compared with commercial cationic reagent (PEI, 25kDa) and liposome. We used a luciferase reporter gene to monitor the biodistribution properties of the emulsion vehicles and to assess the patterns of gene expression in real-time. We found that, while PEI and liposome showed relatively low level of gene expression, emulsion-based bioluminescence showed very stable and efficient gene expression profiles over 8 days. These data demonstrated the feasibility of lipid emulsion as in vivo gene carrier, which could be useful in optimizing the gene therapy approaches for treatment of liver-specific diseases.
