Facile spheroid forming method using thermo-sensitive hydrogel based on glycol chitosan |
초록 |
An in vitro cell culture process to obtain transplantable cells is required for tissue engineering or cell therapy. A two-dimensional (2-D) monolayer culture method has been a standard to obtain transplantable cells. However, in vivo system is 3-D and present differences from 2-D in protein expression, cell proliferation, extracellular matrix, and metabolic function. This may be one of the major reasons for limiting the therapeutic efficacy of transplanted cells. Thus one of the thrusts in the field of cell therapy is to develop better 3-D cell culture method in vitro that would mimic in vivo system. The purpose of this study was to investigate a facile, consistent, and cost-effective 3-D cell culture method in vitro. To that end, thermo-sensitive hydrogel that have reversible sol-gel transition was synthesized and was coated on cell culture plate for 3-D cell culture.
