SYNTHESIS OF DUAL RESPONSIVE HYDROGEL VIA DIELS-ALDER CLICK REACTION FOR THE CONTROLLED DELIVERY OF DOXORUBICIN |

A high drug loading capacity hydrogel for doxorubicin delivery was prepared efficiently by Diels-Alder click chemistry. First, poly(ethylene glycol)-block-poly[styrene-alt-(maleic anhydride)] (PEG-b-PSMA) block copolymers were prepared by the RAFT polymerization. The anhydride groups bearing PSMA moiety were subsequently functionalized with furfuryl amine and hydrazine in a one-pot reaction. Doxorubicin (DOX) was further covalent linked to the block via pH-sensitive hydrazone bond. The Diels-Alder reactions of the furan pendant groups with a disulfide-containing cross-linker resulted in cross-linked hydrogels. The hydrogel loaded DOX showed a high drug loading capacity and efficiency of 15.4% and 85%, respectively. The hydrogel was stable under physiological condition whereas decross-linking took place in reductive acidic environment of tumors leading doxorubicin released rapidly. |
