We developed a novel in situ chitosan-poly(ethylene oxide) (PEO) hydrogel via mechanism of Michael type addition reaction. The chitosan-PEO hydrogel was visually observed after both gelation and hydration. Rheological analysis was performed to detect gelation time, and the results indicated that gelation was completed within 10 minafter mixing the liquid solutions. An extent of hydrogel swelling and the morphologies of its surface and cross sections after dehydration process were analyzed by micro-balance measurement and scanning electron microscopy observation, respectively. After suggesting a mechanism of hydrogel formation via Michael type addition reaction, we analyzed in detail chemistry of chitosan acrylation based on the results of FTIR, XPS and NMR, which is an important indicator later in induction of chitosan-PEO hydrogel formation. While acrylation of chitosan was chemically observed from the new acrylatepeaks from the results of XPS, FTIR and NMR by comparing those of theuntreated chitosan peaks, physical observation was performed by measurement of relative viscosities of chitosan and acrylated chitosan. Further biological analysesfor biomedical applications such as cellular interaction and proliferation are currently under progress. |
