Thermal Stabilization of Carbonic Anhydrase via Cell-display on Corynebacterium for Efficient Capture of Carbon Dioxide |

We introduce a robust whole-cell platform for the efficient hydration and mineralization of carbon dioxide with easy production of stabilized CA by genetically displaying CA inside the crystalline protein structure on bacterial surface. The highest hydration activity of the displayed CA is shown in Porin B with a doublet of GGGGS as a spacer. Hydration activity of the whole-cell catalyst compared with initial activity are maintained about 90 % at 37 oC for one month, 60 % at 50 oC for 2 weeks, and 40 % at 60 oC for 24 h, which are about 4-6 times higher than non-displayed CA. There is no significant destabilization at room temperature for one month. Our studies indicate that the most important point for the thermal stabilization of CA is structural integrity of the cell display platform. |
