제목 Degradation behaviors and release profiles of PLGA devices induced by hydrophilic/hydrophobic drugs and glycolide monomer
초록 In order to investigate the degradation behaviors of PLGA device induced by hydrophilic/hydrophobic drugs and glycolide monomer, as an additive, the conventional wafers were fabricated by direct compression method. 5-fluorouracil (5-FU) as a hydrophilic model drug and dexamethasone as a hydrophobic model drug were incorporated into PLGA wafers. These model drugs were chosen in that they could easily detected by the simple HPLC analyses. In vitro drug release studies were performed in phosphate buffered saline solution (PBS, pH 7.4) at 37℃ under sink condition. From the result of in vitro release studies, the hydrophilic drug release (%) was increased in proportion to drug loading amount and glycolide monomer. However, the hydrophobic drug release (%) was relatively decreased with the increase of drug loading amount. Moreover, the degradation behavior of the hydrophobic drug-loaded wafers showed faster degradation rate in case of less amounts drug-loaded wafers. In conclusion, these degradation behaviors of PLGA were dependant on drugs, including other factors such as degradation conditions, and presence of additives.
