NONOates-Loaded Calcium Carbonate Mineralized Nanoparticles for Nitric Oxide-Mediated Cancer Chemotherapy |

Nitric oxide (NO) has been known to have potential as an adjuvant to cancer chemotherapy. Recently, mineralized nanoparticles based on block copolymer templates have shown efficient loading potential and delivery ability of therapeutic agents. Herein, we aim to develop a NONOates-loaded mineralized nanoparticle (NONOates-MNP) which can deliver nitric oxide intracellularly and act as adjuvant to cancer chemotherapy. The NONOates-MNP was prepared by CaCO3 mineralization using NONOates-loaded poly(ethylene glycol-b-polyethyleneimine) (PEG-b-PEI) block copolymer as templates. CaCO3 core phase of the MNP would effectively protect NONOates at physiological pH. However, at acidic pH of endosome, NONOates would be exposed to an aqueous environment as CaCO3 core phase degrades and release NO molecules at the cytoplasm. Therefore, NONOates-loaded MNP can be used as a novel intracellular NO delivery system for nitric oxide-mediated cancer chemotherapy. |
