Mineralized Robust Nanoparticles for Enhanced Intracellular Nitric Oxide Delivery |

Nitric oxide (NO) has attracted a growing interest because it can express the apoptotic activity for various cancers. At a high concentration level of NO, it can inhibit tumor growth. Herein, we selected S-nitrosoglutathione (GSNO) as an NO precursor. GSNO-loaded calcium carbonate-mineralized nanoparticles (GSNO-MNPs) were prepared by sequentially adding the aqueous solutions of GSNO and CO32- ions to an aqueous mixture of the anionic block copolymers (PEG-Poly(L-aspartic acid)) and Ca2+ ions. The stability of GSNO shielded within the crystalline CaCO3 core was greatly enhanced. The GSNO-MNPs triggered NO release at endosomal pH and an intracellular ascorbic acid level. In vitro cell experiments demonstrated that NO release by the GSNO-MNPs efficiently improved therapeutic activity of doxorubicin. |
