Early detection of cancer diagnosis is essential for accurate diagnosis and surgery. In this study, we designed gadolinium-based T1 contrast agent (CA) for cancer diagnosis. 1,4,7,10-tetraazacyclodecane-1,4,7,10-tetraacetic acid (DOTA) was conjugated to diamino linker and with saccharose which has many hydroxy groups to improve the water proton density around Gd3+ ion by hydrogen bonding. 1H NMR spectrum showed successful synthesis of CA. Also, xylenol orange method and ICP-MS results showed that Gd3+ tightly chelated with CA. In vitro paramagnetic properties of CA showed relatively high intensity compared to Gadovist. Also, we obtained an enhanced contrast intensity comprated to Gadovist at tumor site. These results support that our CA can be used as a cancer diagnostic T1-CA for MRI. |
