A series of amphiphilic graft copolymer, poly(2-hydroxyethyl L-aspartamide) (PHEA) grafted with L-lactic acid oligomers (LAs) (PHEA-g-LA) with various degree of substitution (DS) of LA, was synthesized. Their self-assembled nanostructures showed structural transformation from spherical micelles to ultrasmall unilamellar vesicles according to the increase of DS. By differing of the aggregate preparation method, the vesicles of different size, ultrasamll nanovesicles less than 50 nm, nanovesicles about 150 nm, and microvesicles about 5 μm in diameter, were prepared. Vesicles loaded with hydrophilic anticancer drug doxorubicinhydrochloride were prepared and in vitro loading and release behavior was monitored. They showed effective intracellular delivery and therapeutic effect in the in vitro cell study. |
